Actually, we were just talking about this the other day in my office (we goof off a lot) after somebody brought in some boxed DVD sets of Friends. My top 5 are: Get Smart, I Dream of Jeannie, Happy Days (the first two seasons only), Frasier and Seinfeld.
Overall, I think the 1960s had some great comedies. Apart from my five listed above, I loved The Beverly Hillbillies and The Munsters, as well. MASH was good too when the 1970s came along.
Batman (the 1960s TV series) is my favorite. When I saw it as a kid, I didn't think of it as a comedy, but now when I watch it on DVD, I love it. I also was a Get Smart fan. I hope Steve Carrell's movie inspires today's moviegoers to watch the TV series (think of The Craw and the cone of silence). What else would make my list? The Benny Hill Show, All in the Family and Frasier.
That's an arguable list even for me because I would be just as happy adding "Curb Your Enthusiasm," "Entourage" and "Arrested Development" to the list and taking out any of the others...

Have only seen Seinfeld 3-4 times. Sanford and Son was tough to leave off (Gerry Shandling was a writer there) as was Cheers and Mash.
Can anyone understand the popularity of the Honeymooners? I utterly despised Jackie Gleason and would immediately change the channel if that show was on. Many critics have called it the greatest comedy ever.

Latter favorites Cheers, The Cosby Show, Married with Children and Nightcourt.
All great shows. I would like to add Family Guy, Trailer Park Boys (from Canada, please youtube it) the Kids in the Hall, and godbless Nessie for mentioning WKRP. Mad about You had its moments, so did Family Ties.
In no particular order I would say Simpsons (D'oh), Cheers (Norm!), Seinfeld (So Played), Mash (Incoming Wounded) and Southpark. All brilliant in their own way, but there are so many more great shows. And I'm always good to go with an Andy Richter production. One of these days your show will make it Andy. One of these days.
Fawlty Towers and Monty Python are obvious. What about Dave Allen? Get Smart was also good. It is hard to get it down to five. There is also Hancock'd Hslf Hour, which did have a TV version. Spike Milligan also did some TC stuff.
OK Betting, what about O'Reilly show? That surely qualifies as comic relief for the slow lane crowd. Its funnier than the Simpsons, which is also not mentioned above. Simpsons cartoon characters however are more realistic than O'Reilly.

WKRP definitely had its moments. Fraser? Never clicked with me. House MD has some of the wittiest and sharpest writing, but it's more of a drama.
For anyone who hasn't seen Arrested Development, it's a must-see.
